Responsibilities
This role is for a lab-based Research Scientist with knowledge and experience in transcriptomics / proteomics. This is a hands-on laboratory-based role with focus on effective delivery to support various clinical and preclinical projects across multiple diseases, species and drug modalities.

Qualifications
Essential for the role:

This role will be based in the MPS team at the Early Cardiovascular, Renal and Metabolism (CVRM) department in our dynamic R&D site in Gothenburg, Sweden. The MPS team establishes and applies state-of-the-art MPS platforms to support the CVRM portfolio with emerging therapeutic area needs and new drug modalities. Our mission is to improve translatability from in vitro models to humans and accelerate drug discovery and development.
What you’ll do:
You will execute and provide input into the practical delivery of MPS studies and support the development, characterization, and optimization of new models.

Responsibilities
Take part in the design, conduct, interpretation and reporting of studies with advanced cell models and MPS.
Carry out various downstream analyses from the cell culture samples with versatile in-vitro analytical methods such as, but not limited to, ELISA’s, qPCR, staining and imaging.
Join further model development activities to support drug projects with MPS data for resolving mechanisms of action, target identification, and candidate drug evaluation.
Effectively communicate and collaborate across the Early CVRM and with the key stakeholders in other functions.

Qualifications
Essential for the role:


Master’s degree + 2 years of relevant experience.
Background in Pharmacy, Biology, Biotechnology, Biochemistry, Medical Biosciences, Bioengineering or similar
Strong hands-on cell culture experience
Experience with primary cells or hPSCs
Versatile in-vitro analytical methods (e.g., qPCR, staining and high-content imaging, plate-based assays e.g., ELISAs)
Independent study design, laboratory work, and data analysis
Data visualization and statistical analysis
Excellent interpersonal skills and excellent communication skills in English, both verbal and in writing
Strong ability to engage and collaborate across boundaries with a problem-solving attitude and focus on delivery

Pharmaceutical Sciences (PharmSci) is responsible for the design and development of synthetic routes, preclinical and phase I/II formulations, encompassing both small molecules and new modalities such as mRNA and oligonucleotides, across the whole range of AstraZeneca's therapeutic areas.

Within Early Product Development and Manufacture (EPDM) in Gothenburg, Sweden, the manufacturing unit is accountable for delivering a vast portfolio of early investigational medicinal products. The role
We are now seeking a new member to join our fantastic team, based at AstraZeneca's dynamic R&D site in Gothenburg, Sweden. Within the EPDM manufacturing unit, in the GMP (Good Manufacturing Practice) arena, you will make an important contribution in delivering clinical results in the development of new medicines.

This role is focused on environmental monitoring of the multipurpose GMP facilities and support to the manufacturing within the field of microbiology/hygiene. Therefore, knowledge within microbiology is a requirement for this role. In addition, experience of GMP purified water/water for injection systems is a merit, as accountabilities within these areas are included.

The role

Proven experience with human PSC biology and differentiation to neural cell types
Experience with primary neuronal cultures
Experience in developing biochemical and cell-based assays in primary and PSC-derived neurons, introducing agents by viral transduction or transfection.
Proficiency in cellular and molecular biology techniques, including qPCR, ELISA, Western blotting, Flow cytometry, routine immunofluorescence and IHC staining
Perform in vitro studies to investigate the effects of CRISPR-based gene editing on gene and protein expression and cellular readouts.
Effectively collaborate within a matrixed research team to facilitate the success of projects. Maintain complete experimental records.


The following skills are highly desirable:
Experience with CRISPR gene editing, clonal cell line generation, and NGS library prep
Expertise with liquid handling automation technologies
Experience of setting up cell based pharmacological assays

* Hands-on lab-based experience in utilizing advanced molecular biology RNA-sequencing, DNA-sequencing or Epigenetic sequencing techniques.

Accountabilities and responsibilities

* Accountable for delivery of human target validation in relevant models by experimentally demonstrating presence and functional implication of the target in relevant disease mechanisms, mechanistic understanding of target and disease
* Accountable to identify and develop PoM biomarkers, Decision Making and other Exploratory Biomarkers as required for each target.

Minimum Requirements - Education and Experience
Education

* PhD or equivalent with a strong experimental drive and background in basic science related to respiratory research.
* Post-doc experience or otherwise relevant scientific research experience.
* Experience in pharmaceutical R&D.
* Strong publication record.

Experience and skills

* Broad understanding of several technologies and methodologies along with excellent scientific laboratory skills and ability to experimentally address key scientific questions, including Cell culture: cell lines and primary cells, Nucleic acid isolation, RT-qPCR, ELISA, MSD assays, FACS
* Understanding of the Respiratory therapy area and disease biology, including molecular pathways and mechanisms related to respiratory diseases.
* Excellent skills in hypothesis forming and testing, able to draw conclusions and suggest next steps based on the experimental data.

Responsibilities
The Laboratory Project Coordinator will be collaborating with the Lab team to ensure the work processes are kept according to standards set in the ISO 17025 accreditation and will report to the Sweden COVID-19 Assessment Laboratory Head. Onboarding new personal
Ensuring all staff are documented and compliant regarding mandatory training

Supporting Lab Head regarding quality and SHE issue reporting

As member of the PCR Sweden team, ensure issues and changes to routines are communicated both to and from the lab team
Point of contact for the Assessment Centers for ad hoc questions eg. regarding sampling kits, transport of samples between lab and assessment center, daily lab capacity etc.
Holding regular meetings with Health Care Providers (HCP) providing opportunity for feedback on the process

What you will do:

In this role, you will apply high throughput methods and strategies to projects sought at identifying novel oligonucleotide based therapies.

With your significant expertise within the molecular biology field you will actively be involved in the planning and execution of scientific projects centered around oligonucleotide therapy. You will apply cutting edge technologies and methodologies to establish a high efficiency qPCR oligonucleotide screening capability in the NGS & Transcriptomics team. As well as design and execute NGS experiments to explore the science of selected oligonucleotide; including - primary and secondary mechanistic impacts and off target effects.

We also rely you to:

* Apply this HT-qPCR capability to the activity screening of oligonucleotide libraries to discover novel hits.
* Apply NGS technologies to identify on-target and off-target transcripts.
* Build strong collaborative relationships with the project investigators, key partners, project bioinformaticians and data scientists to ensure the smooth running of projects.

* Candidates should have a Master degree.

The roles
You will be based in Gothenburg Sweden, working alongside our stem cell scientists, disease areas specialists and together with our NGS and PGE team. Using your strong knowledge and experience in cell-based assay development and/or NGS and CRISPR technology you will have the opportunity to work with cutting edge cellular models of disease. This includes culture and differentiation of induced pluripotent stem cells (iPSCs) as a platform for developing disease relevant cellular models to validate target hypothesis.
