Requirements:

We are currently recruiting for a Biobank Scientist in the AZ Gothenburg Biobank group within the global Precision Medicine and Biosamples organization. This position will provide the opportunity to work in a dynamic and enthusiastic environment whilst supplying a critical service to drug discovery projects. The work is highly collaborative with the team having a collective responsibility for the overall delivery.
The role is focused on supporting drug discovery projects within R&D Gothenburg by maintaining high quality handling and storage of HBS and support projects with requested samples in a timely manner.

As a Biobank Research Scientist you will be expected to:
• Receive and register incoming HBS
• Process requests for HBS to customers, including using automated liquid handler

Production of pharmaceutical tablets is moving from a batch-oriented to a continuous flow. Consequently the analysis needed for quality acceptance must be adapted for immediate results. Impurity analysis has still not been adapted for the new setup and for some products also content uniformity is lacking an analytical method.


The aim of this project is to develop and evaluate methodology for direct measurement on tablets by DART-MS or MSMS, and set up a flow for data processing by machine learning methods.


The project is a collaboration between Mass Spectrometry, Machine learning and Material Science experts and will give the student a broad knowledge in several key disciplines in late state drug development. Interest for analytical chemistry and multivariate techniques is needed.



DART-MS is a widely used, nondestructive technique for identifying substances in forensics, security and food industry, but it has so far been rarely used for quality attributes in drug production.  The reason has not been lack in sensitivity but failing precision (ca 10%) as data interpretation has focused only on the measured component. The proposal in this project is to use the full spectrum from a Quadrupole-Time of Flight mass spectrometer (QTOF-MS) and use a trained machine learning algorithm to use the rest of the spectrum as internal standard.  In this way we hope to be able to acquire both the impurity and content analysis in only one run that takes just one second, using no solvents or sample preparation.

I dina arbetsuppgifter ingår framförallt att handlägga kunduppdrag med analyser på GC/MS och LC/MSMS, primärt med standardiserade metoder men även forsknings- och utvecklingsarbete.

Dialogen med våra uppdragsgivare är viktig för att kunna identifiera och förstå kundens egentliga behov. Förutom egna kundkontakter ser vi gärna att du tar initiativ till att vidareutveckla vårt kunderbjudande inom kemiområdet. Vidare ingår rådgivning och stöttning i kundkontakter inom OEKO-TEX.

Vem är du?
Du är analytisk kemist, kemiingenjör eller högskoleingenjör med inriktning analytisk kemi. Yrkeserfarenhet inom analytisk kemi och masspektrometri är ett krav.

För att trivas med arbetsuppgifterna bör du vara flexibel, noggrann, kvalitetsmedveten och resultatinriktad samt ha förmåga att driva arbetet självständigt och strukturerat. What you will do

In this role, you will combine experimental work with digital technologies to gain an understanding of critical material attributes linked to physical particle properties. What You'll do
In this role you will be responsible for design and characterisation of formulations for the delivery of nucleic acids. This will involve working with advanced drug delivery platforms such as lipid nanoparticles and working in close collaboration with peers in the advanced drug delivery group. You will work on improving our end-to-end formulation design, characterisation and testing capabilities. This is a hands-on, lab-based position where you will work with different teams, so you should be a good communicator. Essential for the role

* PhD degree in pharmaceutical sciences, biology, chemistry, or a related discipline
* Experience in formulating colloidal products such as lipid nanoparticles, polymeric delivery vehicles, other nanoparticle technologies
* Strong technical knowledge of nucleic acid-based drug delivery technologies
* Experience in analytical instrumentations used to characterize various formulations (particle size, zeta potential, pKa determination etc.)
* Practical skills with cell-based in vitro assays to assess the performance of nanomedicines
* A working knowledge of CRISPR-related biology
* Hands on flow cytometry
* Understanding of principles, applications and management of Safety Health and Environment (SHE)

Who are you?

Sustainability and material transition is important to you. Technology is a big interest, and you are motivated by practical experimental work. Thesis work: Analytical Characterization of mRNA Lipid Nano Particles Drug Product

Modified mRNA represents a promising new class of therapeutic drug substance. Recently, their impact has been clearly demonstrated through the development of vaccine treatments for Covid-19. However, vaccines are only one class of mRNA therapies and numerous pharmaceutical companies are working on other aligned approaches [1-3]. The number of drug products based on Lipid Nano Particles (LNP) is increasing and applications for different therapies is under intense development. 

To improve successful delivery of these new and novel therapies, it is important to develop methods for the characterization and stability assessment of mRNA drug products.  The mRNA drug substances are highly charged and the intact full-length mRNA consists of a distribution of length variants which has to be handled. Moreover, the therapeutic mRNA is constructed with various modified nucleotides. Altogether, this set an interesting challenge on the analytical method capabilities.

Since the successful application of mRNA technology is relatively new, regulatory guidelines for mRNA quality tests are still evolving. A first draft guideline is now available and several of the methods for purity testing are based on ion pair liquid chromatography separations with UV-detection. [4-5]. 
The focus of this project will be to set up and explore the new ion pair liquid chromatography methods for purity testing of mRNA as described in the first draft guideline.

Background: Biochemical assays are critical in several stages of drug discovery, including high-throughput screening (HTS), lead optimization, and studies of structure-activity-relationship (SAR) and mode-of-action (MoA). A large proportion of drug targets are proteins for which the only known biological function is ligand binding. Such targets require biochemical assays that can measure the interaction between the protein target and a ligand. Often the protein target is expressed as a fusion protein with an affinity tag that can be indirectly “labelled” by a fluorophore-conjugated antibody or other labelled affinity reagent. While this strategy is often successful, it is resource intensive to produce the many protein variants that may need to be tested and there is a significant cost for the labelled affinity reagents. An alternative approach is to directly conjugate a fluorophore to the protein target of interest using a bioorthogonal chemical reaction resulting in a protein target covalently bound to a fluorophore while maintaining its biological activity. Examples of such reactions are amine- or thiol-coupling using N-hydroxysuccinimide- or maleimide-activated fluorophores, respectively. This research is aimed at exploring bioorthogonal protein labelling for biochemical assays and establish a generic labelling protocol for biochemical time-resolved FRET assays.
Hypothesis: Covalently attaching fluorophores to protein targets will result in robust biochemical FRET assays and reduce resource requirements for assay development.
Purpose: Generate fluorophore-conjugated proteins for use in fluorescence applications such as biochemical binding assays to simplify and streamline assay development.

Qualifications

Hands-on experience with library synthesis and automated synthesis is highly desirable.
Expertise in theoretical and practical organic synthesis, retrosynthesis, compound route design, purification and analytical sciences.
Expertise in liquid handling technologies is a plus.
Knowledge of targeted protein degradation (PROTACs, molecular glues) is highly desirable.
Advanced knowledge of modern chemical literature, chemical technologies and applying new ideas to project delivery.
A track record of driving innovation in chemistry related sciences.

Om rollen

Vem är du?

Du har en bakgrund inom utveckling av biobaserade material, gärna från ett kemiskt perspektiv inom fiber- och polymerteknologi. Specialistkunskaper inom cellulosa/polysackarid-kemi relaterat till upplösning, derivatisering och tvärbindning är ett starkt plus. Även erfarenhet av fluidreologi och materialkarakterisering är fördelaktigt. Du motiveras av praktiskt experimentellt arbete samt industrinära och tillämpad forskning. The role
As a Quality Advisor, you will work with early product development manufacturing and packaging teams and be responsible for Development Quality release of internally or externally manufactured Drug Product and Investigational Medicinal Product for clinical trials. You are expected to assure that activities and processes across the development chain are delivered to appropriate standards to meet regulatory expectations and assure product integrity and acceptability.

You will develop and maintain a knowledge of the quality regulations and guidelines applicable to your area and a deep scientific understanding of the products that you are supporting. In your role you will have interactions with internal and external stakeholders.

Typical Accountabilities:
Responsible for quality Release of manufactured Drug Product, Investigational Medicinal Product for clinical trials
Providing quality input to Manufacturing, Packaging, Labelling & Distribution activities
Providing proactive quality advice to support the development, implementation and continuous improvement of GMP quality systems across the Clinical Supply functions and support implementation of agreed global standards
Support the pharmaceutical development functions with investigations of deviations, changes and other quality and compliance decisions in the development projects
Providing quality advice and approval of documentation related to equipments, facilities, IT systems and instruments e.g. maintenance, calibration, validation and change control
Provide appropriate Quality Assurance input to business improvement projects
Operate as part of a global organisation with an integrated mindset to create an organisation with common processes and ways of working.


Qualifications
Minimum requirements – Education and Experience
MSc in pharmacy or engineering (specializing in Pharmaceuticals) with at least 3 years’ experience of working within a pharmaceutical GMP environment
Broad understanding of Quality Systems and GMP and related activities to Investigational Medicinal Product (IMP)
Comprehensive understanding of the pharmaceutical/drug development process
Good communicator with experience of interacting effectively across interfaces
Understanding of Project Management processes
Fluent in written and spoken English

Preferred requirements
Experience preferably within a pharmaceutical manufacturing organization

We propose an opportunity to work closely with our scientists to investigate several key steps in the analytical method development for metabolite profiling of antisense oligonucleotides in biological samples, including sample preparation, separation, and detection. Data processing methods will also be evaluated by comparing a few different softwares. High resolution mass spectrometry as well as radioactivity detection will be used.  Results of this study are expected to contribute significantly to the development of the best-in-practise bioanalytical and data processing approach for biotransformation studies in oligonucleotide platform.

A focus for AstraZeneca Gothenburg is to develop inhaled drugs for the treatment of respiratory diseases such as asthma. A critical domain in this work is to produce active pharmaceutical ingredients (API) particles with appropriate properties e.g. size and morphology, where spray drying is one of efficient formulation platforms. To this end, understanding the relationship between material properties, processing conditions, and product characteristics of spray drying process is an essential component.

Responsibilities
This project aims to characterize the droplet distribution and correlate their characteristics to spray dried particles.

Responsibilities include using advanced particle sizing techniques, e.g. laser diffraction, to measure the size distribution of droplets produced by different atomization nozzles at different conditions e.g. air-to-liquid ratios. This will be accompanied by characterising spray dried powder properties using e.g. scanning electron microscope, in order to build the relationship between material properties, processing conditions and product characteristics. 

If it is of interest, student will also have the option to perform CFD simulations of the atomization process and validate the simulations with experimental data

The work is to be conducted from January to June 2022, or as agreed with the student.

Are you a master’s student looking for thesis work with start spring 2022? Are you interested in the pharmaceutical industry? 

Responsibilities
AstraZeneca offers 30 and 60 credit thesis work within different areas of the company.  They are looking for motivated master students passionate about science and willing to learn a broad range of new techniques.



Qualifications


Currently enrolled as a student within a technical, scientific, IT or other relevant field
A strong interest in pharmaceuticals or the pharmaceutical industry
Open and comprehensive scientific mindset.
Written and oral communication skills in English
Ability to work collaboratively across boundaries and problem solve in the absence of complete information.
